We investigated the effects of long-term intake of wheat bran hemicelluloses (WBH) or guar gum on blood glucose, lipid levels, and abdominal fat accumulation in high-fat-fed mice. Eighteen male C57BL/6J mice were divided into three groups with similar mean body weights and were fed either a: high-fat control diet,high-fat diet containing WBH or guar gum, corresponding to 5% of total dietary fiber for 22 weeks. Adipose tissues were weighed, and plasma total cholesterol, triglyceride and glucose levels were measured. The expression levels of genes involved in cholesterol and fatty acid metabolism in the liver and abdominal fat were measured by quantitative PCR. There were no statistically significant differences in food intake and body weight gain among these groups. Plasma total cholesterol, abdominal fat accumulation (epididymal fat, perirenal fat and retroperitoneal fat) and leptin mRNA of epididymal fat were significantly lower in the WBH group compared with the control group. WBH feeding did not affect liver Hmgcr or Cyp7a1 mRNA expression. Plasma glucose levels and leptin mRNA of epididymal fat were significantly lower in the guar gum group compared with the control group. Brown adipose tissue was significantly increased in the guar gum group compared with the control group. WBH and guar gum may have beneficial effects on a high-fat diet lifestyle and its related diseases.
